Due to this, NGI is now planning to start running the spatial transcriptomics method which enables spatially resolved RNA expression data to be collected from tissue sections. This method uses kits sold under the brand name Visium to be purchased from 10x genomics as they currently are the only provider of these kits.

II.2.4) Description of the procurement:

Part of NGI Stockholms mission as a state-of-the-art facility is making novel techniques available to the research community. Due to this, NGI is now planning to start running the spatial transcriptomics, enabling global assessment of RNA expression in a spatially resolved manner along the area of a tissue section. Briefly, RNA molecules expressed at thousands of spatially defined coordinates in the tissue are subject to cDNA syntesis using oligonucleotides carrying coordinate specific sequence motifs. It is known which sequence motifs correspond to which coordinate and this enables spatially resolved RNA expression levels to be determined after sequencing the synthesized cDNA. This method uses kits sold under the brand name ‘Visium Spatial’ to be purchased from 10x genomics as they currently are the only provider of a solution enabling transciptome-wide, non-targeted spatial transciptomics.

Explanation:

The Visium spatial kits from 10x genomics are intended to be introduced in the operations at NGI Stockholm in a pilot phase to investigate the utility of this technology developed by the company. There are other companies offering products for spatial transcriptomics inference but their products offer targeted assays targeting a subset of distinct RNA. This stands in contrast to the Visium spatial solution by 10X genomics as Visium enables global expression inference any poly-adenlylated mRNA. The reagent kits required are solely provided by 10x genomics as they are protected by patents (US Pat. No 10,030,261, US Pat No 9,593,365, US Pat No 9,371,598, US Pat No 10,308,982, US Pat No 9,868,979 and US Pat No 9,879,313) and no alternatives are thus available on the market.
